Two dirt bacteria just wiped out human tumors in mice—no T cells, no B cells, no immune system required.
Proteus mirabilis (A-gyo, from soil) + Rhodopseudomonas palustris (UN-gyo, photosynthetic) injected IV at 3:97 ratio. They colonize hypoxic tumor zones, then A-gyo explodes to 99:1 inside the mass within 24h.
Mechanism is brutal: local cytokine spike → fibrin deposition + platelet aggregation → selective thrombosis → tumor vasculature collapses. A-gyo elongates into filaments when exposed to fumarate/lactate, penetrates cancer cells, forms biofilms, secretes cytolysins. Iron gets stripped via bacterial siderophores. Result: widespread necrosis.
100% complete response in BALB/c-nu/nu, SCID, NOD-SCID mice with Colon26 tumors. Human xenografts (HT29, SKOV3, BxPC3) vanished. Orthotopic pancreatic model: pancreas weight + CA19-9 marker returned to baseline.
Safety: no cytokine-release syndrome at therapeutic double-dose. Single dose >7.8×10⁹ CFU/ml triggers lethal cytokine storm (fixable with dexamethasone or dose splitting). Bacteria stay imipenem-sensitive, clearable on demand. 150-day survivors showed no organ damage.
Critical detail: ratio matters. Wrong inoculum composition kills mice in 48h.
This isn't immune priming—it's direct vascular sabotage + oncolysis. Earlier 2023 Advanced Science paper showed the same consortium worked in immunocompetent models (colorectal, sarcoma, metastatic lung, TNBC), but the 2025 Nature Biomedical Engineering study proves adaptive immunity is optional.
Led by Eijiro Miyako (JAIST), collab with Daiichi Sankyo + Tsukuba. Startup + Phase 1 trial targeted in 6 years.
Unknown: whether human tumor vasculature responds identically. But mechanism is elegant—unmodified environmental strains self-regulate outside tumors, go nuclear inside.
